Personality:
While Nathan Drake may be the protagonist of the Uncharted series, Sam Drake possesses a lot of the same traits one would expect to find from a roguish leading man. He is a thief with a heart of gold, a penchant for getting into trouble with authorities, and never misses the opportunity to throw out a few sarcastic and witty comments when the opportunity arises. In fact, if you asked Sam his opinion, he would state as he does in game that he was "always the funny one".
He would probably also say that he was the handsome and charming one, as he certainly doesn't avoid the opportunity to flirt when given the chance. There are multiple times throughout the game where Sam is shown as being playfully amorous, such as when Nathan, Sully, and Sam are trying to figure out the best way to steal a cross from a highly guarded auction. A waitress comes over, and Sam wastes no time in flirting with her despite the potentially dangerous situation. A look into one of Nathan's journal entries also shows that one of the useful phrases Sam insisted on learning in Italian was, "Do you have a boyfriend?" to which Nathan writes, "My brother is an idiot."
So despite being the older brother, Sam has a childish and playful attitude about him. Part of this has to due with the amount of years Sam spent in prison. Because he was locked up in his twenties and was there for thirteen years, Sam is now in a strange state of not having completely matured in the same way that his brother has. He clings to the past, and wants things to be the way they were before he was thrown back in jail while at the same time ignoring that the world, and more importantly, his brother have changed. In this way, Sam is also incredibly stubborn and selfish, and goes to extreme lengths to try and get what he wants. We see this when he first reunites with Nate. Instead of accepting the life his brother has made for himself, he wants him to join him in picking up where they left off for the hunt of Avery's lost treasure. He even sprouts what turns out to be an elaborate lie in order to guilt Nathan into joining him, and when confronted later he states that the reason he did it was because the treasure "was always theirs."
Furthermore, Sam can have trouble expressing his emotions. This comes from a tumultuous childhood in which his father abandoned Nate and Sam at a Catholic orphanage after their mother passed away. Sam and Nathan were left to survive on their own, so for Sam, the whole word revolves around them. He has trouble trusting people outside his known circle of contacts, even if they've proven loyal and okay by Nathan. This is seen when Nate and Sam are waiting for Sully to get them into the Italian auction. As they wait, Sam constantly questions Sully's ability and loyalty to them despite Nate trying to convince him otherwise.
In addition, Sam can be blinded by his ambitions and his greed. Like any thief, he thrives on the idea of the prospect of treasure, and the thrill of the hunt and adventure fuels his motivations. Even after lying to his brother and getting into a potentially life threatening situation, Sam insists on trying to get to Avery's treasure despite the chance to escape unscathed. While trying to convince Nate, Elena, and Sully to go after the treasure, Sully states that he "just doesn't know when to quit." Elena also adds that Avery was "hellbent on keeping the treasure. No matter the cost to the others around him," pointedly insinuating that Sam's desire to get the treasure is similar to that of the pirate's.
All that being said, Sam is ultimately a kind hearted and loyal man who would do most anything for those he holds dear to him--especially his brother. As a teenager, Sam raised Nathan by himself, and often looked after him with gestures that ranged from as small as giving him his coat on a cold night, to putting himself between a gun and his brother to try and calm an older woman. Sure, a life of crime may not be the best life to lead for a child, but they survived, and after the whole ordeal with Avery is over, Sam even sneaks a few ancient coins into his sister-in-law's pockets to make sure that they're both taken care of financially.
Setting: At first Sam will likely be very confused as to what’s happening, and won’t really trust what people tell him or the natives here. But once he starts getting used to the idea, he’s more likely to start exploring the worlds for any treasures/just to learn about the different places, and be more accepting to the circumstances.
Abilities/Skills:
Fighting: Sam is a capable fighter who can hold his own fairly well in hand to hand combat situations. Like his brother, his fighting style tends to lean more towards a brawling style and he’s capable of landing a few good punches and knocking somebody out if his enemy is unarmed.
Guns: Sam is capable of using a variety of guns and has shown prowess in different types such as pistols and automatic rifles.
Thievery: Nathan isn’t the only one who picked up a few things off the streets. Like his brother, Sam demonstrates a knowledge of different skills related to thievery including pick pocketing, lock picking, as well as planning bigger heists/cons in collaboration with Nate or Sully. Nathan also credits Sam as being the best pick pocket between Sully and himself.
Stealth: One has to know how to move quietly when one is trying to steal lost artifacts or remain undetected by guards. Sam, like his brother, is a master at it.
Strength/durability: During his time in prison, Sam made sure to keep in shape. He is capable of scaling tall buildings or cliffsides without any harnesses or little equipment, moving large objects, and usually ends up with little more than a few bruises when he falls on his ass too many times.
Languages: Like his brother, Sam is capable of speaking and reading in multiple languages. The ones he is capable of speaking/reading in canon are: Spanish, Latin, a little bit of Italian, and a little bit of Portuquese.
Knowledge of history: Sam is a history nerd and is often described as being an expert on Avery and other pirates. He’s often quick to throw out facts or explanations when prompted, and enjoys doing so.

-Improvisation: Sam is capable of planning big heists, but when things go unexpectedly sour he is also capable of coming up with plans on the spot to get out of tight situations or move around obstacles that might be blocking his way.
-Intelligence/Experience: Sam is capable of helping his partners solve puzzles or clues and never hesitates to provide his own thoughts or advice on a situation when needed.
-Smooth talker: Sam is often seen talking his way out of dangerous situations, even when a gun is pointed at his face. It also helps with the ladies.

-Women: He enjoys the company of women and can sometimes get blind sighted by them.
-Treasure/Gold/Adventure: While Sam enjoys the thrill of adventure, there’s nothing that can compare to a huge payout in the form of ancient pirate treasure. It doesn’t help that Nathan already has some ancient civilizations under his belt as well.
-Adrenaline hunter: Sam wouldn’t be in this business if he didn’t enjoy the adrenaline of it all. Even after the Avery job is finished, he’s quick to offer his help to Sully for his next job.
